Ascorbate is required by prolyl and lysyl hydroxylase to hydroxylate proline and lysine residues in procollagen. Without it the triple helix is unstable and poorly secreted — the biochemical basis of scurvy's connective tissue signs. Hydroxylysine residues also anchor the cross-links that give mature collagen its tensile strength.
Accumulated macromolecular damage is among the recognised hallmarks of biological aging, which is why antioxidant nutrients are studied in this context. Supplementation trials have not shown that antioxidants slow human aging.
