Vitamin C and the collagen triple helix

Ascorbate is required by prolyl and lysyl hydroxylase to hydroxylate proline and lysine residues in procollagen. Hydroxylysine residues also anchor the covalent cross-links that give mature collagen its tensile strength — so vitamin C influences both collagen quantity and quality. Skin holds relatively high ascorbate concentrations.

Biotin, stated honestly

NIH ODS says hair, skin and nail claims for biotin 'are supported, at best, by only a few case reports and small studies.' Benefit is documented in genuine deficiency, which is rare. Biotin supports normal keratin-related metabolism; strong evidence for cosmetic benefit in people who are not deficient is lacking.

Zinc's enzymatic breadth

Zinc is required for the catalytic activity of hundreds of enzymes, which is why deficiency shows up in skin and healing before it shows up anywhere obvious.

Safety, screening & contraindications

  • IMPORTANT — biotin interferes with laboratory tests. Supplemental biotin distorts biotin-streptavidin immunoassays, producing falsely high or falsely low results. The FDA has received adverse event reports of falsely low troponin, including one patient death after a missed heart attack. Tell every clinician and laboratory that you receive biotin, and hold biotin before thyroid, hormone or cardiac testing per your lab's guidance. This also affects TSH, T4, hCG and vitamin D assays.
  • Zinc: adult upper limit 40 mg/day; chronic high intake impairs copper absorption and interacts with quinolone and tetracycline antibiotics.
  • High-dose vitamin C: caution with oxalate kidney stones, hemochromatosis, renal impairment and G6PD deficiency.
